8664 Keystone

Keystone 8664 is a membrane-anchored DNA-binding domain (MADD) that allows proteins to attach to a DNA strand and control gene activity. It is a novel protein-DNA interaction domain found as part of the transcription factor family of proteins. Its primary role is to facilitate the binding of transcription factors to specific DNA target sites and mediate the transduction of regulatory signals from the cell's transcriptional machinery. It is composed of two conserved elements: a helix-turn-helix (HTH) motif and a second amphipathic helix for the attachment of the protein to the DNA. By combining these two conserved elements, Keystone 8664 is able to bind to DNA in a manner that is both highly sequence-specific and flexible. In this way, it can act as an adaptable regulator, allowing transcription factors to respond to specific DNA sequences and respond appropriately to changing conditions.
